I have been accepted and enrolled into the MSc. Physics (International track) programme. I need to submit my admission letter to the visa office in India as part of my documentation. However, my admission letter needs to have the language of instruction of the programme (English) stated in it, as a requirement of the student visa. The University of Hamburg's admission letter does not mention the language of instruction. Can the university issue an official document, stating the language of instruction of my programme?

That is correct. It is not necessary to state the language of instruction in the admission letter, as the degree programme for which you have been admitted is explicitly stated in the admission letter.
The regulations specific to each degree programme at the Universität Hamburg (including the language of instruction) are officially published on our website.
You will find the information regarding the language of instruction in the "Subject-specific Regulations" ("Fachspezifische Bestimmungen" or "FSB") for your degree program:

"Zu § 1 Absatz 1:(1) Der englischsprachige Studiengang Physics (M.Sc.) hat ein forschungsorientiertes Profil."(page 2).
So usually a document for an individual student stating the language instruction isn't necessary, since the regulations for our degree programs are public information.
However, if the responsible institution is not satisfied with this document linked above and insists on an individual document for you indicating the language of instruction, you have to contact the Accademic Office of the Department of Biology at the Unviersität Hamburg, as the departments are responsible for such subject-specific documentation.
